ABSTRACT

Objective:To explore the advantages and disadvantages of frozen section versus rapid paraffin section in the evaluations of donor organ.Methods:Five cases of donor liver and 8 cases of discarded donor kidney were collected from 2017 to 2021.Tissues were harvested and prepared by frozen section, rapid paraffin section and normal paraffin section.After hematoxylin-eosin (H&E) staining, the specimens of donor kidney/liver were evaluated by differential histopathological structures and donor quality scoring system.Results:Rapid paraffin section was similar to normal paraffin section in reflecting the proportion of glomerulosclerosis (18.6%±22.3%), arteriolar hyaline degeneration (43.7%±23.8%) and arteriolar stenosis (47.9%±29%). The proportion of glomerulosclerosis (0.8%±2.2%), arteriolar hyaline degeneration (4.9%±7.4%) and arteriolar stenosis (5.3%±7.5%) were lower in frozen sections than those in rapid paraffin sections.The diagnoses of hydropic degeneration and necrosis in donor liver were more accurate in rapid paraffin section.Conclusions:Rapid paraffin section is superior to frozen section in observing histopathological changes under microscope.Scoring of donor organ is more precise according to rapid paraffin section.

ABSTRACT

Objective To investigate the value of bone marrow imprint in the diagnosis of plasma cell myeloma and the remission rate after chemotherapy.Methods Bone marrow aspiration,bone marrow imprint,and bone marrow biopsy of plasma cell myeloma with 128 patients were collected.The bone marrow biopsy was used as the standard.Bone marrow imprint was compared to bone marrow aspiration in bone marrow nucleated cells quantity,infiltration degree,and diagnostic coincidence rate.Sensitivity,specificity,positive predictive value,and Youden index were evaluated.Results The results of bone marrow imprint showed that the number of nucleated cells was better than that of bone marrow aspiration (P < 0.05),and was similar to that of bone marrow biopsy (P > 0.05).Bone marrow biopsy combined with CD38 (+) showed the degree of infiltration of bone marrow as the standard,and the bone marrow imprint was better than that of bone marrow aspiration (P < 0.05),and was similar to that of bone marrow biopsy (P >0.05).To evaluate the compliance rate of bone marrow infiltration in bone marrow imprint:In phase Ⅰ,the compliance rate of bone marrow aspiration was 95.83% (23/24) and bone marrow imprint of 91.67% (22/24),the difference was not statistically significant (P > 0.05);In phase Ⅱ,bone marrow imprint coincidence rate was 92.86% (65/70),which was significantly higher than that of bone marrow aspiration in 65.71% (46/70) (P <0.05);In phase Ⅲ,bone marrow imprint coincidence rate was 79.41% (27/34),which was significantly higher than that of bone marrow aspiration in 44.12% (15/34) (P < 0.05).The other parameters,such as sensitivity,specificity,positive predictive value,and Youden index were better than those of bone marrow aspiration.Conclusions Bone marrow imprint has characteristics that both bone marrow aspiration and bone marrow biopsy have.It has obvious advantages in diagnosis of plasma cell myeloma and monitoring the remission after chemotherapy.

ABSTRACT

We propose a double layered (intradural and epidural patch) duraplasty that utilizes Lyoplant and Duraseal. We examined a 47-year-old woman after decompression for thoracic ossification of posterior longitudinal ligament was performed in another hospital. On postoperative day 7, she complained of weakness in both legs. Postoperative magnetic resonance imaging (MRI) showed cerebrospinal fluid (CSF) collection with cord compression. In the operative field, we found 2 large dural defects on the ventral dura mater. We performed a conventional fat graft with fibrin glue. However, the patient exhibited neurologic deterioration, and a postoperative MRI again showed CSF collection. We performed dorsal midline durotomy and inserted a intradural and epidural Lyoplant patch. She immediately experienced diminishing back pain postoperatively. Her visual analog scale and motor power improved markedly. Postoperative MRIs performed at 2 and 16 months showed no spinal cord compression or CSF leakage to the epidural space. We describe a new technique for double layered duraplasty. Although we do not recommend this technique for all dural repairs, double-layered duraplasty may be useful for repairing large inaccessible dural tears in cases of persistent CSF leakage refractory to conventional management.

ABSTRACT

Objective To develop an antigen retrieval method for detection of human mammaglobin ( hMAM) immuno-histochemcal staining in old paraffin-embedded specimens .Methods The tissue sections in test group were put into dis-tilled water after deparaffinization and then moved into citric acid buffer ( pH 3.5) for 10-15 min.The other two meth-ods,microwave method and high pressure cooker method ,were compared as control groups at the same time .Finally, immu-nohistochemistry SP method was used to check the antibody in the sections .Results The color appearance in the test group (pH 3.5 citric solution) was better than that of microwave oven and high pressure cooker groups .In the test group, tissue sections were not easily cast off from the slices .Conclusion In this study,we have established a new and simple antigen retrieval method which will contribute to immunohistochemistry technology .

ABSTRACT

Histotechnology is concerned with processing and preparing of the body tissue in such a manner as to enable a satisfactory study of it. Section cutting is an integral part of histology and histopathology. It is an art by itself requiring skill and knowledge on the part of technician or the person who needs to do section cutting. In the routine method of preparing paraffin sections, it is often encountered by the presence of artefacts like fine wrinkles or folds. Attempts have been made to remove the wrinkles by floating the sections in the warm water bath. However this method has not been able to remove all the wrinkles from the sections. We have designed a simple and reliable method, in which the paraffin sections were floated over the ethyl alcohol diluted with water (1:15) before they were placed in the water bath. Through this method, we have been able to get the wrinkle free sections of superior quality. The advantage of our method is that, it is easy to prepare the dilute alcohol and is cost effective. This method can be used by the histology and pathology technicians and the researchers.


Histotecnología se refiere a la elaboración y preparación de los tejidos del cuerpo de tal forma que permitan un estudio satisfactorio de éste. El corte seccionado es una parte integral de la histología y la histopatología. Es un arte por sí mismo que requiere habilidad y conocimiento por parte del técnico o la persona que tiene que hacer el corte de la sección. En el método de rutina de la preparación de las secciones de parafina se encuentra a menudo la presencia de artefactos como arrugas finas o pliegues. Se han hecho intentos para eliminar las arrugas por flotación de las secciones en el baño de agua tibia. Sin embargo, este método no ha sido capaz de eliminar todas las arrugas de las secciones. Hemos diseñado un método sencillo y fiable, en el que las secciones de parafina fueron colocadas por flotación sobre alcohol etílico diluido con agua (1:15) antes de ser colocado en el baño de agua. A través de este método, hemos sido capaces de obtener secciones sin arrugas de calidad superior. La ventaja de nuestro método es que, es fácil de preparar el alcohol diluido y es rentable. Este método puede ser utilizado por los técnicos de histología, patología e investigadores.

ABSTRACT

Microtome is a mechanical instrument used to cutbiological specimens into very thin segments formicroscopic examination. Biological specimens can bepresented in many ways. But more often, thesespecimens are embedded in paraffin wax blocks andthe commonest way of sectioning these specimens canbe achieved by the microtome. The earliest form of themicrotome enabled free hand sectioning of fresh orfixed material using a sharp razor. Modern microtomesare precision instruments designed to cut uniformlythin sections of a variety of materials for detailedmicroscopic examination. Central to the production ofgood sections is the microtome knife. Microtomyvirtually begins and ends with a sharp, blemish-freecutting edge. The introduction of disposable blades hasmade easier the production of good quality, thinsections, but they are often unsatisfactory forsectioning harder tissues, especially bone. A sharp knifeedge free from imperfections is essential for theproduction of good sections. Since many types ofmicrotomes are commercially available in the market,choosing the right microtome is essential forproducing the best result as required. A classificationis proposed that unifies and organizes the variousmicrotomes based on the mode of operation.
